目的:探讨冠心病患者血浆白细胞介素18(IL18)水平是否与基质金属蛋白酶-22和-29(MMP-22和MMP-29)的表达水平相关,以及此类患者血浆中MMP-22及MMP-29水平是否升高。创新点:首次证实在动脉粥样硬化过程中炎症反应可能促进MMP-22及MMP-29的表达,IL18是冠心病患者中调控MMP表达的炎症因子之一。方法:通过免疫印迹分析检测IL18对人体动脉平滑肌细胞MMP-22的表达;通过Mann Whitney检验对来自于194例冠心病患者和68例对照组的前瞻性研究进行分析;通过logistic回归分析冠心病的独立风险因素;通过Pearson相关性分析IL18和MMP-22的表达水平与冠状动脉Gensini积分的相关性;通过多变量Pearson相关性分析血浆MMP-22水平与超敏C反应蛋白(hs-CRP)及甘油三酯水平的相关性;通过Spearman相关性分析血浆IL18水平与MMP-22和MMP-29的相关性。结论:冠心病患者血浆IL18水平升高可能导致此类患者血浆MMP-22和MMP-29水平升高。IL18、MMP-22和MMP-29可能是冠心病的生物标记物和独立风险因素。
Objective: To investigate whether plasma interleukin 18 (IL18) levels are correlated with the expressions of MMP-22 and MMP-29 and MMP-22 and MMP-29 in patients with coronary heart disease (CHD) -29 level is rising. Innovative point: For the first time, it is confirmed that inflammation may promote the expression of MMP-22 and MMP-29 during atherosclerosis. IL18 is one of the inflammatory factors that regulate MMP expression in patients with coronary heart disease. METHODS: The expression of IL-18 on human arterial smooth muscle cells (MMPs) was detected by Western blotting. The prospective study of 194 coronary artery disease patients and 68 control subjects was performed by Mann Whitney test. The logistic regression analysis of coronary heart disease Independent risk factors; Pearson correlation analysis of IL18 and MMP-22 expression levels and coronary artery Gensini score correlation; multivariate Pearson correlation analysis of plasma MMP-22 levels and high-sensitivity C-reactive protein (hs-CRP) and Triglyceride levels; correlation of plasma IL-18 levels with MMP-22 and MMP-29 was analyzed by Spearman’s correlation. Conclusion: Elevated plasma levels of IL-18 in patients with coronary heart disease may result in elevated plasma levels of MMP-22 and MMP-29 in these patients. IL18, MMP-22 and MMP-29 may be biomarkers of coronary heart disease and an independent risk factor.
