目的:STAT(signal transducer and activators of transcription)是一种重要的细胞质转录因子,在恶性肿瘤的发生、增殖和凋亡中发挥着重要作用。本研究旨在探讨STAT3单核苷酸多态性(single nucleotide polymorphisms,SNP)与胃癌生物学行为的关系。方法:运用SYBR GREEN PCR熔解曲线法检测93例胃癌患者的STAT3SNP。结果:随机抽取18例胃癌患者以基因测序检测STAT3SNP,结果与SYBR GREEN PCR熔解曲线法完全相同。含有G等位基因的CG+GG基因型相对于CC基因型,脉管侵犯以及神经侵犯差异性显著(P<0.001)。结论:SYBR GREEN PCR熔解曲线法可以作为检测STAT3SNP方法之一。STAT3SNP与胃癌的某些生物学行为相关,该位点为G等位基因提示肿瘤脉管侵犯、神经侵犯高风险。
Objective: STAT (signal transducer and activators of transcription) is an important cytoplasmic transcription factor that plays an important role in the carcinogenesis, proliferation and apoptosis of malignant tumors. This study aimed to investigate the relationship between STAT3 single nucleotide polymorphisms (SNPs) and the biological behavior of gastric cancer. Methods: SYBR GREEN PCR melting curve was used to detect STAT3 SNP in 93 patients with gastric cancer. Results: Stat3SNP was detected by gene sequencing in 18 gastric cancer patients randomly. The result was identical with that of SYBR GREEN PCR. CG + GG genotypes with G allele had significant differences (P <0.001) in vessel invasion and neuroinvasion over CC genotypes. Conclusion: SYBR GREEN PCR melting curve method can be used as one of the methods to detect STAT3 SNP. STAT3SNP is associated with some biological behaviors of gastric cancer. This site suggests that the G allele implicates tumor vascular invasion and high risk of nerve invasion.
