背景:作者先前的研究表明,在实验性胃溃疡自愈期间,胃窦粘膜β-内啡肽(beta-endorphin,β-EP)细胞,胰岛β-EP细胞均发生变化,但前脑beta-EP神经元是否发生变化还不清楚。目的:观察大鼠实验性胃溃疡期间前脑β-EP神经元的变化。设计:本研究采用双盲、随机、对照方法。地点和材料:本研究在河北北方学院组织学与胚胎学教研室进行,研究对象为成年雄性Wistar大鼠(180~230g),购自军事医学科学院动物中心,随机分为实验性胃溃疡组(溃疡组)19只、盐水对照组(盐水组)15只和正常对照组(正常组)4只。干预:溃疡组,在胃前壁近胃窦处,注入0.01mL冰醋酸至胃粘膜下层,造成实验性胃溃疡;盐水组,模拟假手术,注射等量生理盐水代替冰醋酸;正常组,为正常大鼠,不加任何处理。在实验性胃溃疡手术后1,4,10和23d分4批心脏灌流取脑,每批均包括溃疡组和对照组。干预者为作者本人。主要观察指标:前脑β-EP神经元在大鼠实验性胃溃疡术后1,4,10及23d溃疡自愈过程中的分布、形态和数量变化。结果:正常大鼠前脑β-EP神经元主要分布于弓状核及其周围,在大脑皮层和杏仁复合体也显示了少量β-EP弱阳性神经元。溃疡术后1d,大脑皮层纹状皮质β-EP阳性胞体罕见,纤维亦不明显,溃疡组β-EP神经元数与盐水组相比无差异;杏仁复合体、下丘脑各核区β
BACKGROUND: Previous studies by the authors showed that during the healing of experimental gastric ulcer, the changes of β-endorphin (β-EP) cells and islet β-EP cells in gastric antral mucosa were all changed. However, Whether EP neurons change is unclear. Objective: To observe the changes of β-EP neurons in forebrain during experimental gastric ulcer in rats. Design: This study used a double-blind, randomized, controlled approach. Location and Materials: The study was conducted at the Department of Histology and Embryology, North China University of Hebei. The study was performed on adult male Wistar rats (180-230g) purchased from the Animal Center of the Academy of Military Medical Sciences and randomly divided into experimental gastric ulcer group (ulcer 19), saline control group (saline group) 15 and normal control group (normal group) 4. Intervention: ulcer group, in the anterior gastric wall near the antrum, injection of 0.01mL glacial acetic acid to the gastric mucosa, resulting in experimental gastric ulcer; saline group, simulated sham operation, injection of normal saline instead of glacial acetic acid; normal group, Normal rats, without any treatment. At 4, 10, and 23 days after experimental gastric ulcer surgery, four batches of cardiac perfusion were used to take the brain. Each batch consisted of ulcer group and control group. The interventionist is the author himself. MAIN OUTCOME MEASURES: The distribution, morphology and quantity changes of forebrain β-EP neurons in healing process of ulcer in rats with experimental gastric ulcer at 1, 4, 10 and 23 days after operation. Results: The β-EP neurons in the normal rat forebrain mainly distributed in and around the arcuate nucleus. A small amount of β-EP weak positive neurons were also found in the cerebral cortex and almond complex. In the ulcer group, β-EP positive cell bodies in the cortex of the cerebral cortex were rare and the fibers were not obvious at 1 day after operation. The number of β-EP neurons in the ulcer group was not different from that in the saline group.
