目的:建立测定异黑成熟颗粒中2种有效成分田蓟苷和甘草酸含量的方法。方法:采用高效液相色谱法,Diamonsil C18(4.6 mm×250 mm,5μm)色谱柱,乙腈-0.5%甲酸(27∶73)为流动相,检测波长324 nm,柱温35℃,流速1.0 mL.min-1。Diamonsil C18(4.6 mm×250 mm,5μm)色谱柱,甲醇-0.2 mol.L-1酸铵-冰醋酸(67∶33∶1)为流动相,检测波长250 nm,柱温30℃,流速1.0 mL.min-1。结果:田蓟苷和甘草酸分别在1.14~114μg(r=0.999 9)和20.6~206μg(r=0.999 7)线性关系良好;平均回收率分别为98.26%(RSD 2.29%),99.39%(RSD 1.86%)。结论:该方法简便可行,重复性好,可用于异黑成熟颗粒中田蓟苷和甘草酸含量测定。
Objective: To establish a method for the determination of the contents of thymosin and glycyrrhizic acid in two kinds of active ingredients of Heihe Mature Granules. Methods: The mobile phase consisted of Diamonsil C18 (4.6 mm × 250 mm, 5 μm), acetonitrile-0.5% formic acid (27:73), detection wavelength 324 nm, column temperature 35 ℃ and flow rate 1.0 mL .min-1. The mobile phase was Diamonsil C18 (4.6 mm × 250 mm, 5 μm), methanol-0.2 mol·L-1 ammonium acetate-glacial acetic acid (67:33:1) mL.min-1. Results: The calibration curves were linear in the range of 1.14-114 μg (r = 0.999 9) and 20.6-206 μg (r = 0.999 7) respectively. The average recoveries were 98.26% (RSD 2.29%) and 99.39% (RSD 1.86%). Conclusion: The method is simple, feasible and reproducible. It can be used to determine the contents of thymidine and glycyrrhizic acid in mature black particles.
