为了建立早期和晚期原发性乳腺癌的裸鼠模型,将15例乳腺原位癌和31例浸润性癌移植于155只裸鼠皮下。新鲜的癌组织被制备成细胞粗悬液和组织小块,分别植入到裸鼠两背侧皮下,同时植入一粒17-β-雌二醇缓释丸于右侧皮下。在10个月观察期中,原位癌终未长出,光镜下可见移植物仍维持原位癌的结构特征,未见浸润性发展。31例浸润性癌中仅1例明显生长并被反复传代。该原发瘤的病理诊断是多中心去分化导管癌,在裸鼠体内生长的移植瘤经鉴定不表达雌、孕激素受体,p53蛋白阴性,cerbB-2蛋白阳性,DNA倍性为近四倍体。经多次传代后,移植瘤仍能维持原发瘤的各种特征,因此这一模型的建立为晚期乳癌的研究提供了一个新的工具。早期乳癌的裸鼠模型虽未建成,但为今后进一步的研究积累了重要的实验资料
To establish a nude mouse model of early and late primary breast cancer, 15 cases of breast carcinoma in situ and 31 cases of invasive carcinoma were transplanted subcutaneously in 155 nude mice. Fresh cancer tissue was prepared into a crude cell suspension and tissue pieces, which were implanted subcutaneously on both dorsal sides of nude mice and a 17-β-estradiol sustained-release pellet was implanted on the right side of the skin. In the 10-month observation period, carcinoma in situ did not grow at all, and under the light microscope, the graft still maintained the structural features of carcinoma in situ, and no invasive development was seen. Of the 31 cases of invasive carcinoma, only 1 was significantly developed and was repeatedly passaged. The pathological diagnosis of the primary tumor is a multicenter dedifferentiated ductal carcinoma. The transplanted tumors grown in nude mice do not express the estrogen and progesterone receptors. The p53 protein is negative, the cerbB-2 protein is positive, and the DNA ploidy is nearly four. Ploidy. After multiple passages, the transplanted tumor can still maintain various characteristics of the primary tumor, so the establishment of this model provides a new tool for the study of advanced breast cancer. Although the nude mouse model of early breast cancer has not been completed, it has accumulated important experimental data for further research in the future.
