目的 :探讨维甲酸对胎鼠面突外胚间充质细胞的作用和CyclinD1变化之间的关系。方法 :原代培养GD12 1 5(妊娠第 12天第 15小时 )胎鼠面突的外胚间充质细胞 ,原代培养细胞后 ,观察维甲酸处理前后细胞生长曲线的变化 ,采用免疫组织化学和图像分析仪对维甲酸处理后CyclinD1蛋白的表达变化进行定量分析。结果 :维甲酸处理后 ,外胚间充质细胞的生长受到抑制 ,正常的外胚间充质细胞中 ,CyclinD1的表达量较高 ,维甲酸处理后 ,其表达量显著降低。结论 :维甲酸所引起的细胞生长抑制与CyclinD1的表达降低之间密切相关 ,外胚间充质细胞的正常分裂需要CyclinD1的正常表达
OBJECTIVE: To investigate the effect of retinoic acid on the changes of embryonic rat embryo-derived embryonic mesenchymal cells and CyclinD1. Methods: Primary culture of GD12 1 5 (15th day of pregnancy on the twelfth day) of fetal rat embryonic mesenchymal cells was performed. Primary cultured cells were cultured and observed. The changes of cell growth curve before and after treatment with retinoic acid were observed. Immunohistochemistry And image analyzer quantitative analysis of CyclinD1 protein expression changes after retinoic acid treatment. Results: After treated with retinoic acid, the growth of ectomesenchymal cells was inhibited. In normal ectomesenchymal cells, the expression of CyclinD1 was high, and the expression of CyclinD1 was significantly decreased after treatment with retinoic acid. CONCLUSION: Retinoid-induced inhibition of cell growth is closely related to the decrease of CyclinD1 expression, and the normal division of ectomesenchymal cells requires the normal expression of CyclinD1
