通过光镜观察毛蚶血细胞形态和结构,对其分类进行研究,分析不同类型血细胞吞噬能力,并研究温度对血细胞吞噬功能的影响。结果鉴定出毛蚶血细胞中有3种类型:(1)大透明细胞,大小为(12.97±1.22)μm,核质比为0.311±0.016,所占比例为(46.40±8.73)%;(2)颗粒细胞,大小为(9.98±1.85)μm,核质比0.390±0.058,所占比例为(41.48±8.63)%;(3)小透明细胞,大小为(7.60±1.17)μm,核质比为0.499±0.049,所占比例为(12.12±4.00)%。毛蚶血细胞在血淋巴液中的平均密度为(7.69±0.98)×106 cell/m L。体外吞噬活性试验表明,大透明细胞和颗粒细胞对酵母聚糖的吞噬率显著高于小透明细胞,吞噬率分别为(61.77±3.30)%、(61.32±3.62)%、(16.59±3.88)%。于15、20、25、30、35℃温度下进行毛蚶血细胞的吞噬试验,结果表明温度变化对血细胞吞噬能力产生明显的影响,30℃条件下,血细胞的吞噬活性最强,吞噬率达到(51.49±4.22)%。
Morphology and structure of the blood cells were observed by light microscope, the classification was studied, the phagocytic ability of different types of blood cells was analyzed, and the effect of temperature on hemophagocytic function was also studied. The results showed that there were three types of cells in the hairy alburnus: (1) large transparent cells with a size of (12.97 ± 1.22) μm and a nucleoplasm ratio of 0.311 ± 0.016, accounting for (46.40 ± 8.73)%; (2) The cell size was (9.98 ± 1.85) μm and the ratio of nuclear to cytoplasm was 0.390 ± 0.058, accounting for (41.48 ± 8.63)%. (3) Small clear cells with a size of 7.60 ± 1.17 μm and a nuclear to cytoplasm ratio of 0.499 ± 0.049, accounting for (12.12 ± 4.00)%. The average density of hairy blood cells in haemolymph was (7.69 ± 0.98) × 106 cells / m L. In vitro phagocytic activity assay showed that phagocytosis rates of zymosan by large clear cells and granulosa cells were significantly higher than that of small clear cells (61.77 ± 3.30%, 61.32 ± 3.62%, (16.59 ± 3.88)%, . At 15, 20, 25, 30, and 35 ℃, phagocytosis of blood cells was observed. The results showed that the changes of temperature had a significant effect on the phagocytic ability of blood cells. Under 30 ℃, the phagocytic activity of blood cells was the strongest, ± 4.22)%.
