[目的]建立高效液相色谱法测定鱼腥草不同部位中天然槲皮素含量的方法。[方法]利用纤维素酶与超声提取相结合;采用色谱柱为Eclipse XDB-C18(4.6 mm×150 mm,5μm),流动相为乙腈-1%冰乙酸(40∶60),柱温为25℃,流速为0.8 ml/min,检测波长为370nm。[结果]槲皮素在0.1~14.4μg/ml范围内呈良好线性关系(r=0.999 9),鱼腥草根样品的平均回收率为99.2%,RSD为3.9%,茎叶样品的平均回收率为98.8%,RSD为3.8%。[结论]所建立的方法分离效果好,专属性强,操作简便,结果准确,重现性好,适用于鱼腥草中槲皮素含量的测定。
[Objective] To establish a method for the determination of natural quercetin in different parts of Houttuynia cordata with HPLC. [Method] Cellulase was combined with ultrasonic extraction. The column was Eclipse XDB-C18 (4.6 mm × 150 mm, 5 μm). The mobile phase was acetonitrile-1% glacial acetic acid (40:60) ℃, flow rate of 0.8 ml / min, detection wavelength of 370nm. [Result] Quercetin showed a good linearity (r = 0.999 9) in the range of 0.1-14.4μg / ml, the average recovery of root samples of Houttuynia cordata was 99.2% and the RSD was 3.9%. The average recovery of quercetin Was 98.8% with a RSD of 3.8%. [Conclusion] The established method has the advantages of good separation effect, special specificity, simple and convenient operation, accurate result and good reproducibility. It is suitable for the determination of quercetin in Houttuynia cordata Thunb.
