试验分别采用四个纤维用亚麻(Linumusitatissimum)品种7309,948,Belinka和Viking的无菌苗的茎尖为材料游离原生质体。以萌发10天的无菌苗茎尖游离获得的原生质体得率和活性最高,分别达到1.8×10~6/gFW和85.5%。以V-KM为培养基,采用琼脂岛法培养的原生质体,可在培养3天后发生第一次分裂,10天后统计细胞分裂频率为36%,20天后统计植板率达到5.2%。品种7309和Belin-ka再生的愈伤组织接种在均附加0.6mg/L 6-BA和0.1mg/L NAA的B_5-1和MS_3固体培养基上,都有芽苗分化,并分别获得再生植株。品种Viking和948分别仅分化获得了不定根或叶状体。
Four fibrolactam-free protoplasts were obtained from the apices of four sterile fibers of Linumusitatissimum varieties 7309, 948, Belinka and Viking, respectively. The highest protoplast yield and activity were obtained from the apex of germ-free 10-day germination shoots, reaching 1.8 × 10 ~ 6 / g FW and 85.5% respectively. The protoplasts cultured on V-KM medium by agar island method could be the first division after 3 days of culture. The frequency of cell division was 36% after 10 days and 5.2% after 20 days. The regenerated callus of variety 7309 and Belin-ka were inoculated on solid medium B_5-1 and MS_3 with 0.6 mg / L 6-BA and 0.1 mg / L NAA respectively, . Varieties Viking and 948 were only differentiated to obtain adventitious roots or fronds.
