目的评估ProCOUNT方法和双平台ISHAGE方法检测脐带血CD34~+细胞计数的相关性及优缺点。方法采用流式细胞仪ProCOUNT和双平台ISHAGE对228份脐带血进行CD34~+细胞计数,对结果进行配对t检验和Spearman分析。结果 228份脐带血经ProCOUNT方法和双平台ISHAGE方法检测的CD34+绝对计数、CD34~+相对计数、有核细胞数差异均无统计学意义(P>0.05)。2种方法绝对计数具有强相关性(r=0.824,P<0.001),相对计数具有中相关性(r=0.714,P<0.001)。结论 2种方法可在检测CD34~+细胞时相互通用,为脐带血库的检测方案提供更多的选择。
Objective To evaluate the correlation and the advantages and disadvantages of ProCOUNT and dual platform ISHAGE in detecting cord blood CD34 ~ + cell count. Methods Flow cytometry ProCOUNT and dual-platform ISHAGE were used to count CD34 + cells in 228 cord blood. The results were analyzed by paired t-test and Spearman analysis. Results 228 cases of umbilical cord blood by ProCOUNT method and dual platform ISHAGE method detected CD34 + absolute count, CD34 ~ + relative count, the number of nucleated cells was no significant difference (P> 0.05). The absolute counts of the two methods were strongly correlated (r = 0.824, P <0.001), and the relative counts had a moderate correlation (r = 0.714, P <0.001). Conclusion The two methods can be used to detect CD34 + cells, which provides more choices for the detection of cord blood bank.
