作者采用高压液相层析方法定量测定片剂及血样中的茶碱。对象为8例健康成年男性受试者,均给予以下4种茶碱片剂:(1)Sio-phyllin Gyrocaps;(2)茶碱缓释片(Theolair-SR片);(3)实验缓释片;(4)普通片。口服单剂量为500 mg。在服药后一定时间采静脉血样,血浆迅速离心分离,取1ml血样与空白样品(无水茶碱加空白人血浆)对照测定,并绘制标准曲线。以血药浓度的对数对时间作图,用线性回归法计算药物的血浆半衰期。药-时曲线下面积AUC按梯形规则计算。
The authors used high pressure liquid chromatography to quantify theophylline in tablets and blood samples. Subjects: Eight healthy adult male subjects were given the following four kinds of theophylline tablets: (1) Sio-phyllin Gyrocaps; (2) Theophylline sustained release tablets (Theolair-SR tablets); (3) Piece; (4) ordinary piece. A single oral dose of 500 mg. Venous blood samples were collected at a certain time after taking the drug and the plasma was rapidly centrifuged. A 1 ml blood sample and a blank sample (anhydrous theophylline plus blank plasma) were measured and a standard curve was drawn. The logarithm of plasma concentration versus time was plotted and the plasma half-life of the drug was calculated using linear regression. The area under the drug-time curve, AUC, is calculated as a trapezoidal rule.
