miR-155、miR-146a在口腔扁平苔藓患者外周血和组织中的变化与相关性研究

目的 :探讨miRNA(miR-155、miR-146a及miR-146b)在口腔扁平苔藓(oral lichen planus,OLP)患者外周血及病损组织中的表达及其与OLP临床特征的关系,为进一步探讨OLP的发病机制提供参考。方法:采用荧光定量PCR法,以U6基因为内参,分别检测OLP患者外周血及病损组织、健康对照外周血及组织中miR-155、miR-146a及miR-146b的相对含量。采用SPSS17.0软件包对数据进行统计学分析。结果 :荧光定量PCR检测显示,与正常对照组相比,外周血miR-155表达水平在OLP患者中显著增高,病损组织中升高更加明显(组织中miR-155水平是外周血中的7.0倍);而OLP患者外周血中miR-146a、miR-146b含量与对照组相比无显著差异(P=0.218,P=0.229),但OLP患者病损组织中miR-146a表达升高,显著高于对照组(P=0.003),且组织中miR-146a水平是外周血中的5倍。相关性分析结果显示,外周血中miR-155的表达水平与REU计分呈高度正相关(相关系数R=0.887,P<0.01)。结论:OLP患者外周血和组织中miR-155、miR-146a异常变化表明,miR-155、miR-146a可能与OLP的发生与演进有关,OLP可能是以局部炎症为主的慢性炎症性疾病。 OBJECTIVE: To investigate the expression of miRNAs (miR-155, miR-146a and miR-146b) in peripheral blood and lesion tissues of patients with oral lichen planus (OLP) and their relationship with the clinical features of OLP. OLP pathogenesis provide a reference. Methods: The relative content of miR-155, miR-146a and miR-146b in peripheral blood and diseased tissues, healthy controls and peripheral blood and tissues of OLP patients were determined by fluorescence quantitative PCR and U6 gene. SPSS17.0 software package for statistical analysis of the data. Results: Fluorescent quantitative PCR showed that compared with the normal control group, the expression of miR-155 in peripheral blood was significantly increased in OLP patients, and the level of miR-155 in the tissue was more obvious (the level of miR-155 in peripheral blood was 7.0 (P = 0.218, P = 0.229). However, the expression of miR-146a in OLP patients was significantly higher than that in the control group Higher than the control group (P = 0.003), and the level of miR-146a in the tissues was 5 times higher than that in the peripheral blood. Correlation analysis showed that the expression level of miR-155 in peripheral blood was positively correlated with REU score (correlation coefficient R = 0.887, P <0.01). CONCLUSIONS: The abnormal changes of miR-155 and miR-146a in peripheral blood and tissues of OLP patients suggest that miR-155 and miR-146a may be related to the occurrence and progression of OLP. OLP may be a chronic inflammatory disease characterized by local inflammation.
