目的调查了人类白细胞抗原(humanleucocyte antigen, HLA)Cw位点在中国汉族人群中的等位基因分布规律,为进一步研究Cw位点的遗传特征提供背景资料。方法采用序列特异性引物扩增技术对1285名无关个体进行HLA-Cw基因特异性分析,并对其分布规律进行统计学分析。结果共检测到23种HLA-Cw位点的等位基因,其中HLA-Cw*01、*03、*07、*08为主要基因型,其基因频率分别为0 .1529、0 .2385、0 .1747、0 .1004 ,并检出HLA-Cw*12、*14、*15、*16、*17等血清学方法不能鉴定的HLA-Cw基因;统计结果表明HLA-Cw位点的基因分布符合Hardy-Weinberg平衡(χ2=73 .74 ,df=98 ,P>0 .5)。结论本研究结果为中国汉族人群提供了一套较为完整HLA-Cw基因遗传学参数。
Objective To investigate the allele distribution of human leucocyte antigen (HLA) Cw locus in Chinese Han population and provide background information for further study on the genetic characteristics of Cw locus. Methods A total of 1285 unrelated individuals were analyzed for HLA-Cw gene using sequence-specific primer amplification and their distribution was statistically analyzed. Results A total of 23 alleles of HLA-Cw locus were detected, of which HLA-Cw * 01, * 03, * 07 and * 08 were the major genotypes and their gene frequencies were 0.1529,0.2385 and .1747,0. 1004, and detected HLA-Cw gene can not be identified by HLA-Cw * 12, * 14, * 15, * 16, * 17 and other serological methods; statistical results show that HLA-Cw locus gene distribution Comply with Hardy-Weinberg equilibrium (χ2 = 73.74, df = 98, P> 0.5). Conclusion The results of this study provide a complete set of HLA-Cw genetic genetic parameters for Chinese Han population.
