目的观察小半夏汤对化疗性异食癖大鼠多巴胺(DA)合成与代谢的影响,探讨小半夏汤防治化疗性恶心呕吐的作用机制。方法取Wistar大鼠108只,随机分为6组,即正常对照组,小半夏汤正常对照组(1.6g·kg-1),顺铂模型组,昂丹司琼组(2.6 mg·kg-1),小半夏汤大、小剂量组(3.2,1.6 g·kg-1),每组18只,单笼饲养。适应性饲养7 d后,造模前4 d投放高岭土颗粒。正常对照组和顺铂模型组给予等容量蒸馏水,灌胃给药,每天2次,首次给药后1 h顺铂模型组和各治疗组动物腹腔注射6 mg·kg-1顺铂建立大鼠化疗性异食癖模型。正常对照组和小半夏汤正常对照组腹腔注射等容积生理盐水。造模后每12 h称量并记录动物高岭土摄入量。分别于造模后24 h和72 h取材,测定大鼠血清、回肠和延髓DA及其代谢产物3,4-二羟基苯乙酸(DOPAC)含量以及回肠和延髓酪胺酸羟化酶(TH)、单胺氧化酶B(MAOB)水平。结果顺铂模型组大鼠摄食高岭土量显著升高,小半夏汤大、小剂量组均可抑制化疗大鼠摄食高岭土,均可降低化疗大鼠血清、回肠和延髓DA含量以及回肠、延髓TH水平,对DOPAC含量和MAOB水平没有明显影响。结论小半夏汤防治化疗性恶心呕吐作用机制与降低外周与中枢TH水平、抑制DA合成有关。
Objective To observe the effects of Xiaobanxia Decoction on the synthesis and metabolism of dopamine (DA) in rats with chemotherapy-induced pica and to explore the mechanism of Xiaobanxia Decoction in preventing and treating chemotherapy-induced nausea and vomiting. Methods One hundred and eight Wistar rats were randomly divided into 6 groups: normal control group, Xiaobanxia Tang normal control group (1.6g · kg -1), cisplatin model group, ondansetron group (2.6 mg · kg- 1) Xiaobanxia Tang large and small dose group (3.2,1.6 g · kg-1), each group of 18, a single cage feeding. After 7 d of adaptive feeding, 4 days before model making, kaolin granules were placed. Normal control group and cisplatin model group were given equal volume of distilled water, intragastric administration, 2 times a day, 1 h after the first administration cisplatin model group and each treatment group animals intraperitoneal injection of 6 mg · kg -1 cisplatin rat Chemotherapy pica. The normal control group and Xiaobanxia Tang normal control group were injected intraperitoneally with equal volume of normal saline. Weigh and record the animal kaolin intake every 12 h after modeling. The serum, ileum and medulla oblongata and their metabolites 3,4-dihydroxyphenylacetic acid (DOPAC) content and ileum and medullary tyrosine hydroxylase (TH) were determined at 24 h and 72 h after modeling respectively. , Monoamine oxidase B (MAOB) levels. Results The dose of feeding kaolin in cisplatin model group was significantly increased. Xiaobanxia decoction large and small dose groups could inhibit the ingestion of kaolin in the chemotherapy rats, which could decrease the content of DA in serum, ileum and medulla oblongata, and the level of ileum and medulla oblongata , No significant effect on DOPAC content and MAOB level. Conclusion Xiaobanxia Decoction can prevent and treat chemotherapy-induced nausea and vomiting and reduce peripheral and central TH levels and inhibit DA synthesis.
