以开胸冠状动脉结扎法制备兔急性心肌梗死(AMI)模型,并设假手术对照组,各25只。AMI组中12只(随机分组)肌注黄茂注射液,另13只同期注射等量生理盐水,各2周。模型制备成功后6周再次开胸取出心脏,取左室壁心肌组织测定羟脯氨酸含量。结果:AMI组梗死区心肌羟脯氨酸含量明显高于周边区心肌及对照组(均P<0.01),黄芪注射组与生理盐水注射组心肌羟脯氨酸含量无明显差异(P>0.05)。结论;①胶原基质在AMI后左室重构中起一定作用;②该模型确实可靠,心肌羟脯氨酸测定可用来评价AMI模型制作方法成功与否;③黄芪无明显作用。
Rabbit acute myocardial infarction (AMI) model was established by ligation of thoracotomy and sham-operated control group, 25 rats each. In the AMI group, 12 (randomized) intramuscular injections of Huangmao injection and the other 13 rats were injected with equal volume of saline for 2 weeks. 6 weeks after the successful preparation of the model, the heart was reopened and the left ventricle myocardium was used to determine the hydroxyproline content. RESULTS: Myocardial hydroxyproline content in the infarcted area of AMI group was significantly higher than that in the peripheral myocardium and the control group (all P<0.01). There was no significant difference in myocardial hydroxyproline content between the Astragalus injection group and the saline injection group (P> 0.05). Conclusion; 1 Collagen matrix plays a certain role in left ventricular remodeling after AMI; 2 The model is reliable, and myocardial hydroxyproline assay can be used to evaluate the success of the AMI model; 3 Huangqi has no significant effect.
