目的探讨肾炎防衰液对5/6肾切除(5/6nephrectomize,5/6NT)大鼠肾组织纤维粘连蛋白(fibronectin,FN)表达的影响。方法选用SD雄性大鼠,建立5/6NT大鼠肾纤维化模型。随机分为8组:正常组、假手术组、模型组、苯那普利组、尿毒清组和肾炎防衰液低、中、高剂量组。采用免疫组织化学方法,以苯那普利、尿毒清为实验对照,检测肾炎防衰液对肾组织FN表达的影响。结果模型组肾组织FN强阳性表达,在系膜基质和肾小管基底膜出现大量棕黄色颗粒;苯那普利组、尿毒清组FN中等量表达,在系膜基质和肾小管基底膜出现中等量棕黄色颗粒;肾炎防衰液低、中、高剂量组肾组织FN轻度表达明显少于模型组,在系膜基质和肾小管基底膜出现少量棕黄色颗粒,并呈剂量依赖关系。结论肾炎防衰液抑制5/6NT大鼠肾组织FN的表达,可能是该方减缓肾小球硬化和肾纤维化发展的作用机制之一。
Objective To investigate the effect of nephritis anti-aging solution on expression of fibronectin (FN) in renal tissue of 5/6 nephrectomized (5/6NT) rats. Methods Male Sprague Dawley rats were used to establish 5/6 NT rat renal fibrosis model. Randomly divided into 8 groups: normal group, sham operation group, model group, benazepril group, uremic clear group and nephritic anti-aging liquid low, middle and high dose groups. Using immunohistochemistry, Benazepril and Uremicin were used as experimental controls to detect the effect of nephritis anti-failure fluid on renal FN expression. Results In the model group, FN expression was strongly positive in the renal tissue, and a large amount of brownish yellow particles appeared in the mesangial matrix and basement membrane of the renal tubule. Fn was moderately expressed in the benazepril group and the uremic clear group, and the mesangial matrix and the tubular basement membrane appeared moderately. The quantity of FN lightly expressed in kidney tissue of the low, medium, and high dose nephritis group was significantly lower than that of the model group. A small amount of brownish yellow particles appeared in the mesangial matrix and the basement membrane of the tubules in a dose-dependent manner. Conclusion The anti-failure solution of nephritis inhibits the expression of FN in the kidney of 5/6NT rat, which may be one of the mechanisms of this side to slow the development of glomerular sclerosis and renal fibrosis.
