利用水稻的愈伤组织作受体 ,采用 PIG基因枪法 ,首先成功地将 Bt基因 [cry IA(b) ]连同抗除草剂 bar基因导入栽培水稻品种中国 91,选育出纯合稳定的株系 T91系。然后将 T91系与黄淮区主栽品种豫粳 6号杂交 ,并辅以标记基因选择 ,选育出纯合稳定、综合性状优良的转 Bt基因株系 C48。利用农杆菌介导的转化系统将 X a2 1基因转入 C48,根据标记基因测定 ,转基因植株的 PCR和 Southern分析 ,田间抗病虫性鉴定 ,获得了双抗 (螟虫、白叶枯病 )转基因水稻
Using callus of rice as receptor, the Bt gene [cry IA (b)] was successfully introduced into cultivated rice cultivar 91 in China using the PIG gene gun method, and the homozygous stable strain T91 system. Then, T91 line was crossed with Yujing 6, a main cultivar in Huanghuai District, supplemented by marker gene selection, and a transgenic Bt line C48 with stable and comprehensive characters was selected. Xa2 1 gene was transferred into C48 by Agrobacterium-mediated transformation system. According to the marker gene analysis, PCR and Southern analysis of the transgenic plants and identification of the resistance to pests and diseases in the field, a double-resistant (stem borer, bacterial blight) transgene Rice
