目的 :从猪脑中分离神经肽 ,并对其氨基酸组成进行分析。方法 :新鲜猪脑经匀浆后用生理盐水和蒸馏水提取。提取液用SephadexG5 0柱层析纯化。收集液做薄层层析并用茚三酮显色。收集Rf(Removefactor) 0 5 0的组分 ,进行硅胶柱层析。收集液再次做薄层层析和茚三酮显色 ,收集Rf值为 0 5 0的组分 ,冷冻干燥制成猪神经肽。结果 :该肽经日立 83 5 5 0型氨基酸分析仪分析 ,证明其含有Ala和Asp二种氨基酸。结论 :结果表明该肽是一种酸性神经肽
OBJECTIVE: To isolate neuropeptides from porcine brain and to analyze their amino acid composition. Methods: Fresh porcine brain was homogenized and then extracted with normal saline and distilled water. The extract was purified by SephadexG 50 column chromatography. The collected solution was subjected to thin layer chromatography and developed with ninhydrin. The fractions of Rf (Removefactor) 0 5 0 were collected and subjected to silica gel column chromatography. The collected solution was subjected to thin-layer chromatography and ninhydrin coloration again, and the fraction having a Rf value of 0 5 0 was collected and freeze-dried to prepare porcine neuropeptide. Results: The peptide was analyzed by Hitachi 836500 amino acid analyzer and proved to contain two kinds of amino acids, Ala and Asp. Conclusion: The results show that the peptide is an acidic neuropeptide
