通过软琼脂克隆法及有限稀释法,从(Hca-F25/L)细胞系中分离出五株不同转移力的克隆细胞,它们的共同特点是:均沿淋巴系统转移,仅有极少数(1.88%)同时伴有肺转移;五株克隆及其亲代对角蛋白单抗(HK2)均呈现阳性,提示尽管它们分化很低,但仍保持了上皮组织的特性;经染色体分析证实,每一克隆内同一核型的细胞占绝大多数,它们和亲代有相同的四个标记染色体,表明它们之间有“血缘”关系。它们的不同之处表现在:在615小鼠体内均能致瘤(100%),但在淋巴结中的转移率及其转移程度有很大的差别,其中一株高转移细胞系(78.6%—89.4%),三株中转移细胞系(33.3%—66.6%),一株低转移细胞系(15%~15.4%);五株克隆及其亲代的干系核型也各不相同;在细胞形态上和细胞电泳率方面,高、低转移细胞系与亲代细胞系三者之间有明显的差别。本文较确切地证明了 Hca 瘤株在转移表型和核型上的异质性。同时,也补充和发展了 H22/Hca 肿瘤系统。
Using the soft agar cloning method and the limiting dilution method, five strains of cells with different transfer forces were isolated from the (Hca-F25/L) cell line. Their common feature was that they all migrated along the lymphatic system with only a few (1.88). (%) was accompanied by lung metastases; five clones and their parents were positive for keratin monoclonal antibody (HK2), suggesting that despite their very low differentiation, they retained the characteristics of epithelial tissues; chromosomal analysis confirmed each clone. The cells that share the same karyotype account for the vast majority. They have the same four marker chromosomes as the parent, indicating a “blood” relationship between them. Their differences are as follows: in 615 mice can be tumorigenic (100%), but the lymph node metastasis rate and its degree of transfer are very different, in which a highly metastatic cell line (78.6% - 89.4%), three metastatic cell lines (33.3%-66.6%), one low-metastatic cell line (15%-15.4%); five clones and their parental karyotypes are also different; in cell morphology On the electrophoretic rate of cells and cells, there were significant differences between the high and low metastatic cell lines and the parental cell lines.
