本文报道用结肠癌转移淋巴结分离的癌细胞免疫小鼠制得4株单克隆抗体即MC3、5、7、10,杂交瘤细胞已经过150天的连续培养,至今分泌抗体稳定,抗体效价高。能直接用于福尔马林固定石蜡包埋的切片作染色。经免疫组化法对成人的几种癌组织、癌旁组织及胚胎组织中MC相应抗原的定位研究,发现MC相应抗原在癌旁正常组织及胚胎组织表达十分局限,但在结肠癌表达可达100%,而且在染色强度、阳性细胞数等方面均比胃癌、食管癌、肺癌显著,提示MC相应抗原是在结肠癌占优势的一种抗原。
In this study, we report that MC3, 5, 7, and 10 monoclonal antibodies were obtained from mice immunized with cancer cells isolated from metastatic lymph nodes of colon cancer. The hybridoma cells have been continuously cultured for 150 days. The antibodies secreted so far are stable, and the antibody titer is high. . Can be directly used for formalin-fixed paraffin-embedded sections for staining. The immunohistochemistry method was applied to the localization of MC antigens in several cancer tissues, paracancerous tissues, and embryonic tissues of adults. It was found that the expression of MC antigens was very limited in adjacent normal tissues and embryonic tissues, but it was expressed in colon cancer. 100%, and the staining intensity, the number of positive cells, etc. are all significantly higher than that of gastric cancer, esophageal cancer, and lung cancer, suggesting that the corresponding antigen of MC is an antigen dominant in colon cancer.
