笔者建立了来源于人脑颞枕部和顶部的两个胶质母细胞瘤的体外细胞系,目前已分别存活5年,传500余代。 方法与结果 一、细胞系的建立:1.材料来源:TJ899细胞系取材于1例男性在颞枕部胶质瘤患者的手术切除标本。TJ905体外细胞系取材于1例男性顶部胶质瘤患者。二者均经病理组织学诊断为多形性胶质母细胞瘤。2.建系经过:在无菌条件下,用Hanks液漂洗瘤组织,除去血液及肉眼可见的电灼及坏死组织。机械离碎,将组织块接种于25cm~2培养瓶内,加入含15%胎牛血清(FCS)的Way Mouth MAB 87/3(Gibco Lab,USA)培养液,37℃、5%CO_2孵育72小时。大多数组织块均有不同程度细胞移出,胞体较小,1周后多数培养瓶壁已长满细胞。经0.125%胰蛋白酶
