神经诱向生长是神经科学前沿中一个重要研究方向,提取与纯化诱向生长因子更是神经生物化学研究中的一个重要技术课题。本研究采用自然系统凝胶电泳,分离周围神经损伤过程中产生的具有诱向生长活性作用的18KD蛋白,再以等电点聚焦凝胶电泳与神经组织联合培养,揭示了PI为5.2的18KD蛋白具有诱导神经生长的作用。高效液相色谱仪分析提示,通过以上方法可获得较纯的18KD诱向因子。本研究为神经诱向因子的分离,纯化与进一步的蛋白序列分析提供了可行的途径。
Nerve-induced growth is an important frontier in neuroscience. Extracting and purifying induced growth factors is an important technical issue in neurobiochemistry. In this study, natural system gel electrophoresis was used to separate the 18KD protein induced by the growth-inducing activity in the process of peripheral nerve injury, and the co-culture of isoelectric focusing gel electrophoresis and neural tissue revealed that the PI was 5.2 18KD protein has the function of inducing nerve growth. High performance liquid chromatography analysis suggested that the above method can be more pure 18KD induced factor. This study provided a feasible approach for the isolation, purification and further protein sequence analysis of neurotrophic factors.
