目的 :探讨胃癌细胞株MGC80 3在全反式维甲酸 (all rans retinoic ,RA)、佛波酯 (phorbolester,TPA)及二甲基亚砜 (dimethylsulfoxide ,DMSO) 3种诱导剂分别作用后 ,胃癌细胞中间隙连接蛋白 (connexin ,Cx)基因表达变化。方法 :采用比较多重RT -PCR -Southernblot方法检测。结果 :维甲酸、二甲基亚砜对胃癌细胞基因组中Cx43基因有诱导表达作用 ,同时该株细胞本身表达的Cx46在维甲酸及佛波酯作用后明显减弱甚至消失。结论 :Cx43基因在胃癌细胞中有可诱导性 ,这为今后探索维甲酸、二甲基亚砜等对胃癌细胞恶性表型的抑制 ,为胃癌细胞基因组中Cx43基因的表达模式在临床上的应用提供了理论依据和实验基础。
Objective: To investigate the effects of gastric cancer cell line MGC80 3 on the effects of all-trans retinoic (RA), phorbol ester (TPA) and dimethylsulfoxide (DMSO) inducers. Changes in Connexin (Cx) gene expression in cells. Methods : A multiplex RT-PCR-Southern blot assay was used. RESULTS: Retinoic acid and dimethyl sulfoxide induced the expression of Cx43 gene in the genome of gastric cancer cells. At the same time, the Cx46 expressed in the cell itself was significantly weakened or even disappeared after the action of retinoic acid and phorbol ester. Conclusion: The Cx43 gene is inducible in gastric cancer cells. This will be used to explore the inhibition of malignant phenotypes of gastric cancer cells by retinoic acid and dimethyl sulfoxide in the future. It is clinically applicable to the expression pattern of Cx43 gene in the genome of gastric cancer cells. Provides theoretical basis and experimental basis.
