目的探讨新疆汉族T2DM及糖耐量正常(NGT)人群miR-375启动子区部分CpG位点的甲基化状态。方法采用基质辅助激光解析电离时间飞行质谱仪检测100例T2DM患者(T2DM组)和100名NGT者(NGT组)外周血白细胞中miR-375基因启动子区5个CpG位点的甲基化状态,并对结果进行分析。结果 (1)T2DM组平均甲基化水平高于NGT组(10.27%vs 7.24%,P<0.01);(2)T2DM组miR-375基因5个CpG位点的甲基化率均高于NGT组,如CpG_22、CpG_24.25、CpG_26.27,仅CpG_26.27在两组中比较,差异有统计学意义(P<0.01)。结论汉族T2DM患者外周血白细胞中miR-375基因部分启动子区异常甲基化水平可能与T2DM的发生有一定的关系。
Objective To investigate the methylation status of some CpG loci in miR-375 promoter region in T2DM and normal glucose tolerance (NGT) populations of Xinjiang Han nationality. Methods The methylation status of five CpG sites in miR-375 gene promoter region in 100 T2DM patients (T2DM group) and 100 NGT group (NGT group) was detected by matrix-assisted laser desorption / ionization time-of-flight mass spectrometry , And analyze the result. Results (1) The average methylation level of T2DM group was higher than that of NGT group (10.27% vs 7.24%, P <0.01). (2) The methylation rates of five CpG sites of miR-375 gene in T2DM group were higher than those in NGT group Groups such as CpG_22, CpG_24.25, CpG_26.27, CpG_26.27 only showed significant difference between the two groups (P <0.01). Conclusion The abnormal methylation level of some promoter region of miR-375 gene in peripheral blood leukocytes of T2DM patients may be related to the occurrence of T2DM.
