目的研究雌激素低下大鼠模型对更年期综合征的模拟程度,为建立我国“辅助改善更年期综合征的功能学评价方法”提供依据。方法以雌二醇(0.2mg/kg)为阳性物,用大鼠建立雌激素低下模型,测定大鼠尾温和血清雌二醇、卵泡刺激素、黄体生成素、β-内啡肽的浓度,观察这些指标对更年期综合征相应指标的模拟程度,并用不同剂量的大豆异黄酮(50、100、200mg/kg)对该模型进行验证。结果试验期间雌激素低下模型组大鼠尾温显著升高,雌二醇和不同剂量大豆异黄酮均不同程度地降低大鼠尾温。试验结束时,雌激素低下模型组大鼠雌激素水平显著低于对照组、雌二醇组和各剂量大豆异黄酮组;雌激素低下模型组大鼠黄体生成素水平显著升高,雌二醇组和高剂量大豆异黄酮显著降低血清黄体生成素水平;雌激素低下模型组血清卵泡刺激素的水平显著高于其他各组,而β-内啡肽的水平显著低于其他各组。结论雌激素低下大鼠模型能够较好地模拟更年期综合征,大豆异黄酮能够一定程度地改善该模型造成的更年期综合征。
Objective To study the degree of simulation of climacteric syndrome in a rat model of estrogen-induced depression and provide a basis for establishing a functional evaluation method for aiding the improvement of climacteric syndrome in China. Methods The estradiol (0.2 mg / kg) was used as a positive substance and the estrogen-lowering model was established in rats. The rat tail temperature, serum estradiol, follicle stimulating hormone, luteinizing hormone and β-endorphin were measured. Observe these indicators of the corresponding indicators of menopausal syndrome simulation, and with different doses of soy isoflavones (50,100,200 mg / kg) to verify the model. Results The tail temperature of rats with estrogen-induced depression was significantly increased during the experiment. Estradiol and different doses of soy isoflavones decreased the rat tail temperature to some extent. At the end of the experiment, the levels of estrogen in the estrogen-deficient model group were significantly lower than those in the control group, the estradiol group and the different doses of soy isoflavone group; the estrogen-lowering model group rats were significantly elevated in the level of luteinizing hormone, estradiol Group and high-dose soy isoflavones significantly reduced serum luteinizing hormone levels; serum estrogen low-dose group of serum follicle-stimulating hormone levels were significantly higher than the other groups, and β-endorphin levels were significantly lower than the other groups. Conclusion The rat model of estrogen-lowering can better simulate climacteric syndrome. Soy isoflavones can improve the climacteric syndrome induced by this model to a certain extent.
