[目的]通过观察葛根素对去卵巢大鼠和铅中毒大鼠所致的骨质疏松症的治疗效果,为绝经后妇女的骨质疏松症和铅中毒所引起的骨质疏松症的防治提供实验依据。[方法]取5月龄健康雌性大白鼠100只,分成5个实验组(每组20只):(1)正常对照组(Normal);(2)去卵巢模型组(OVX);(3)铅中毒模型组(LPD);(4)葛根素+去卵巢(P+0VX)组;(5)葛根素+铅中毒(P+LPD)组。用葛根素治疗骨质疏松大鼠8周后麻醉取血,处死取其骨。骨组织切片形态观察,对骨和血的钙、磷、铅及血中碱性磷酸酶进行测定。数据进行统计学分析。[结果]在0VX组和LPD组中骨钙、骨磷、血钙及血磷均出现降低(P﹤0.01),血碱性磷酸酶升高(P﹤0.01),骨组织形态呈现骨质疏松的病理改变。P+0VX组的骨代谢生化指标(P﹥0.05)和骨组织形态正常,而P+LPD组的骨代谢生化指标(P﹤0.01)和骨组织形态呈现骨质疏松特征的改变。LPD组和P+LPD组的骨铅和血铅明显高于Normal组(P﹤0.01)。[结论]葛根素对去卵巢大鼠的骨质疏松症有良好治疗效果,而对铅中毒所致的骨质疏松症无效;铅中毒可引起骨质疏松症。
[Objective] To observe the therapeutic effect of puerarin on osteoporosis induced by ovariectomized rats and lead poisoning rats and provide the prevention and treatment of osteoporosis caused by lead poisoning in postmenopausal women Experimental basis. [Methods] 100 healthy female rats of 5 months old were divided into 5 experimental groups (20 in each group): (1) normal control group (2) ovariectomized group (OVX); (3) (4) puerarin + ovary (P + 0VX) group; (5) puerarin + lead poisoning group (P + LPD). Treatment of osteoporosis rats with puerarin 8 weeks after anesthesia to take blood, sacrifice their bone. Bone tissue morphology observation, the bone and blood calcium, phosphorus, lead and blood alkaline phosphatase were measured. Data for statistical analysis. [Results] The levels of osteocalcin, osteophosphatemia, serum calcium and phosphorus in 0VX group and LPD group were decreased (P <0.01), and the level of serum ALP increased (P <0.01). The biochemical markers of bone metabolism (P> 0.05) and the morphology of bone in P + 0VX group were normal, while the biochemical indexes of bone metabolism (P <0.01) and morphological changes of osteoporosis in P + LPD group were changed. Bone lead and blood lead in LPD group and P + LPD group were significantly higher than those in Normal group (P <0.01). [Conclusion] Puerarin has a good therapeutic effect on osteoporosis in ovariectomized rats, but is not effective on osteoporosis caused by lead poisoning. Lead poisoning can cause osteoporosis.
