以桔梗的子叶和下胚轴为外植体,采用离体培养的方法,研究了诱导芽的频率,找出较为理想的外植体类型和相应的培养条件,以期建立高效的桔梗组培再生体系,为桔梗优良株系的繁育和利用提供有效途径。结果表明:桔梗的子叶、下胚轴均可成功诱导出再生芽,其中以子叶诱导率最高,最适合的培养基为MS+1.0 mg·L~(-1) BA+0.5 mg·L~(-1)IAA,在MS+0.5mg·L~(-1)?BA+1.0mg·L~(-1)?IAA的培养基上桔梗再生芽的生长长度最高。因此,桔梗的子叶是最理想的外植体类型,相应的分化培养基组合为MS+1.0mg·L~(-1)BA+0.5mg·L~(-1)?IAA,伸长培养基组合为MS+0.50mg·L~(-1)?BA+1.0mg·L~(-1)?IAA。
Using the cotyledons and hypocotyls of Platycodon as explants, we studied the frequency of induced buds and found the ideal explant types and corresponding culture conditions by using the method of in vitro culture, in order to establish a highly efficient regeneration of Platycodon System, which provides an effective way for the breeding and utilization of good lines of Campanulaceae. The results showed that the regenerated shoots could be successfully induced from the cotyledons and hypocotyls of Campanulaceae, and the most suitable culture medium for cotyledons was MS + 1.0 mg · L -1 BA + 0.5 mg · L ~ (-1) -1) IAA. The regeneration of Campanulaceae shoots had the highest length of growth on MS + 0.5 mg · L -1 BA + 1.0 mg · L -1 IAA medium. Therefore, cotyledon cotyledon is the most ideal explant type, and the corresponding differentiation medium combination is MS + 1.0 mg · L -1 BA + 0.5 mg · L -1 · IAA, elongation medium The combination of MS + 0.50 mg · L -1 BA + 1.0 mg · L -1 IAA.
