目的 研究鸡眼视网膜多巴胺转运蛋白 (dopaminetransporter ,DAT)的分布及其在实验性近视眼形成中的变化及可能作用。方法 孵化第 2天的雏鸡随机分为 4组 ,Ⅰ、Ⅱ、Ⅲ组小鸡单眼分别戴 10D、 2 0D透镜及半透明眼罩 ,IV组为正常对照组。 3周后测量双眼屈光状态及眼轴长度。一侧大腿肌注12 5I β CIT ,二小时后处死小鸡 ,摘出眼球 ,解剖出全层视网膜组织 (Ⅰ及Ⅳ组 )或视网膜神经上皮层与视网膜色素上皮层 (retinalpigmentepithelium ,RPE)分别取下(Ⅱ及Ⅲ组 ) ,以β 计数器分别测定各组织标本多巴胺转运蛋白的放射性计数。结果 RPE组织标本检测到的DAT放射性计数明显高于视网膜神经上皮层。实验眼视网膜组织标本DAT放射性计数大于对照眼。结论 视网膜DAT主要分布在RPE。视网膜DAT可能参与形觉剥夺性近视及透镜诱导性近视的形成。这种研究方法为今后深入研究多巴胺系统与实验性近视眼的关系提供了一条新的途径。
Objective To study the distribution of dopamine transporter (DAT) in corns and its change in experimental myopia and its possible role. Methods Chickens on the second day after hatching were randomly divided into 4 groups. Groups I, II and III were given monocular 10D and 20D lenses and translucent blinds respectively. IV group was normal control group. After 3 weeks, both eyes were measured refractive status and axial length. One side of the thigh muscle 125I β CIT, two hours after the death of the chickens, the eye was removed, dissected full-thickness retinal tissue (Ⅰ and Ⅳ group) or retinal neuroepithelium and retinal pigment epithelium (retinal pigment epithelium, RPE) were removed (Group Ⅱ and Ⅲ). The radioactivity count of dopamine transporter in each tissue was determined by β counter. Results The DAT radioactivity counts in RPE tissue specimens were significantly higher than those in the retinal neuroepithelium. The experimental eye retinal tissue specimens DAT radioactivity count is greater than the control eye. Conclusion Retinal DAT mainly distributes in RPE. Retinal DAT may be involved in the formation of form-deprivation myopia and lens-induced myopia. This research method provides a new way for further study on the relationship between dopamine system and experimental myopia in the future.
