本文用受体放射分析技术,建立人外周血淋巴细胞核T_3、T_4和rT_3受体测定法,并探讨其与甲状腺功能的关系。分离人外周血淋巴细胞采用聚蔗糖-泛影葡胺分层液密度梯度离心法。受体结合试验采用完整细胞分别与~(125)I-T_3、~(125)I-T_4、~(125)I-rT_3温育的方法,用0.0075%TritonX-100 SMT pH7.85溶液分离核,用Scatchard法分析受体结合的解离常数(Kd)和最大结合容量(MBC)。
In this paper, receptor radiometric analysis was used to establish the T_3, T_4 and rT_3 receptor assay of human peripheral blood lymphocytes and its relationship with thyroid function. Peripheral blood lymphocytes were isolated by Ficoll-diatrizoate dextran gradient centrifugation. Receptor binding assay The intact cells were incubated with ~ (125) I-T_3, ~ (125) I-T_4 and ~ (125) I-rT_3 respectively and the nuclei were separated by 0.0075% TritonX-100 SMT pH7.85 The dissociation constants (Kd) and maximal binding capacity (MBC) of the receptor binding were analyzed by the Scatchard method.
