本文在不分离的情况下,研究在70%甲醇提取液中和70%乙醇提取液中分别测定牛黄解毒丸的番泻甙和黄芩甙,并探索二组分薄层层离的条件。结果显示:采用和三氯化铁—氢氧化钠比色法直接测定番泻甙(在490nm 测定),不受干扰,显色稳定时间为10分钟,回收率为95.5%(n=8)。黄芩甙采用内标标准系列工作曲线外推法,在紫外276nm 处测定,方法回收率为98.9%(n=5)。薄层层析条件为硅胶 G 板,二甲基甲酰胺:丙酮为1.5:10预展一次后,作分配薄层层离,正丁醇:醋酸:水为4:1:5,展距:2cm 后,获得二组分的△Rf 达0.3,斑点分离清晰度高,题示物二组分分离获满意结果。本文还检测了二种不同工艺(已灭菌和未灭菌)的牛黄解毒丸二组分含量变化情况,其中黄芩甙测定获得 CV%=0.438%,r=0.9997,发现未灭菌产品含量均稍高。
In this paper, without separation, we studied the senna and jaundice of Niuhuang Jiedu Pills in 70% methanol extract and 70% ethanol extract, and explored the conditions of the two-component thin layer delamination. The results showed that the colorimetric method was used for the direct determination of senna (measured at 490 nm) with ferric chloride-sodium hydroxide without interference. The color stabilization time was 10 minutes and the recovery was 95.5% (n=8). Astragalus membranaceus uses the internal standard standard series of work curve extrapolation method, measured at UV 276 nm, and the method recovery rate is 98.9% (n=5).
