低密度脂蛋白(low-density lopoprotein,LDL)受体是广泛存在于各种组织细胞膜的跨膜糖蛋白。据报道,LDL 受体活性与血浆胆固醇水平及冠心病的发生有明显关系,测定细胞 LDL 受体对动脉粥样硬化发病机理的研究及诊断家族性高胆固醇血症(familialhypercholesterolemia,FH)等具有重要意义。现采用Na~(125)I 标记 LDL,用放射受体测定方法检测人淋巴细胞受体活性,以研究缺血性脑、心血管疾病患者的LDL 受体功能状态及其在发病中的意义。此方法敏感、结果可靠、重复性好,可作为早期发现家族性高胆固醇血症的诊断特异指标。
Low-density lipoprotein (LDL) receptors are transmembrane glycoproteins that are found in a wide variety of cell membranes. It is reported that LDL receptor activity and plasma cholesterol levels and the occurrence of coronary heart disease have a clear relationship between the determination of cellular LDL receptor on the pathogenesis of atherosclerosis and diagnosis of familial hypercholesterolemia (familialhypercholesterolemia, FH) is important significance. We use Na ~ (125) I labeled LDL to measure the activity of human lymphocyte receptor by radioactive receptor assay to study the functional status of LDL receptor and its significance in the pathogenesis of ischemic brain and cardiovascular diseases. Sensitive to this method, the results are reliable, reproducible, and can be used as early detection of familial hypercholesterolemia diagnosis specific indicators.
