采用大鼠颈总动脉插管法及尾动脉体表血压测量法对海洋药物六硝酸甘露醇(mannitol hexani-trate,MH)进行了急性及慢性降压实验,观察了MH对遗传性高血压大鼠及正常大鼠的降压作用。MH[80mg/(kg.d)]经ig连续用药30d后,使遗传性高血压大鼠的血压由用药前的198±12mmHg降至172±6mmHg,平均降低了26±8mmHg,与NS对照组比较差异显著(P<0.05)。同样剂量的MH经ig30d后,使正常大鼠的尾动脉血压平均降低12±7mmHg(P>0.05)。在急性降压实验中,采用颈动脉插管法,MH[80、40/kg.d)]经十二指肠内给药,使正常大鼠的颈总动脉血压降低了14±6mmHg,与NS组比较,差异不显著(P>0.05)。在由乙酰胆碱和组胺诱发的支气管哮喘的模型中,预先igMH(80mg/kg),每日两次,连续用药1w,可使豚鼠的引喘潜伏期延长33±10s,与NS组比较差异显著(P<0.05)。实验结果表明,MH对遗传性高血压大鼠有较温和的降压作用,并对实验性支气管哮喘有一定的预防作用。
Acute and chronic antihypertensive tests were performed on the mannitol hexani-trate (MH) in rats by intubating the common carotid artery of the rat and measuring the blood pressure on the cranial artery surface. MH was observed for hereditary hypertension. Hypotensive effect in rats and normal rats. MH [80mg/(kg.d)] reduced the blood pressure of hereditary hypertensive rats from 198±12 mmHg before treatment to 172±6 mmHg, an average of 26±8 mmHg, after ig administration for 30 days. The difference was significant (P<0.05). After the ig30d of the same dose of MH, the tail artery blood pressure of normal rats was reduced by 12±7mmHg (P>0.05). In acute hypotensive experiments, carotid arterial cannulation was used, and MH [80, 40/kg.d] was administered intraduodenally to reduce the blood pressure of the common carotid artery in normal rats by 14±6 mmHg. Compared with NS group, the difference was not significant (P>0.05). In a model of bronchial asthma induced by acetylcholine and histamine, the guinea pig’s induced asthma latency was prolonged by 33±10 s, which was significantly different from that in the NS group (80 mg/kg) twice a day for 1 hr. P<0.05). The experimental results show that MH has a mild antihypertensive effect on genetically hypertensive rats and has a certain preventive effect on experimental bronchial asthma.
