目的:研究比较柴胡桂枝汤——先合后煎、柴胡桂枝汤——先煎后合、小柴胡汤、桂枝汤方与方间对CCl4引起大鼠急性实验性肝损伤的保护作用。方法:SD大鼠随机分为六组,正常对照组,CCl4模型组,柴胡桂枝汤——先合后煎组,柴胡桂枝汤——先煎后合组,小柴胡汤组,桂枝汤组,分别将各汤剂给SD大鼠灌胃(ig)给药,用CCl4制备急性肝损伤模型,常规生化方法检测血清中丙氨酸氨基转移酶(ALT)、天门冬氨酸氨基转移酶(AST)活性、肝组织中谷胱甘肽过氧化物酶(GSH-PX)和丙二醛(MDA)水平;HE染色,观察肝脏组织的病理变化。结果:CCl4致大鼠急性肝损伤时血清中ALT、AST活性明显升高,肝组织GSH-PX活性显著下降,MDA含量显著升高。中药柴胡桂枝汤——先合后煎、柴胡桂枝汤——先煎后合、小柴胡汤、桂枝汤均可明显降低急性肝损伤大鼠血清ALT,AST活性及肝脏MDA含量,提高肝脏GSH-PX活性。结论:柴胡桂枝汤——先合后煎、柴胡桂枝汤——先煎后合、小柴胡汤、桂枝汤能保肝降酶,抗脂质过氧化,对CCl4所致大鼠急性肝损伤具有明显的保护作用。柴胡桂枝汤——先煎后合中的桂枝汤与小柴胡汤药效间相互作用可能存在“相恶”的类“七情”关系。
Objective: To compare the protective effect of Chaihu Guizhi Decoction - Preconstrip combined decoction, Chaihu Guizhi Decoction - Xianjian Houhe, Xiaochaihu decoction and Guizhi decoction on acute experimental hepatic injury induced by CCl4 in rats. Methods: SD rats were randomly divided into six groups: normal control group, CCl4 model group, Chaihu Guizhi Decoction - Xianhehoujian decoction group, Chaihu Guizhi Decoction - Xianjiangyuxu group, Xiaochaihu decoction group and Guizhi decoction group The rats were given intragastric (ig) administration of each decoction to SD rats, and acute liver injury model was established by CCl4. Serum levels of alanine aminotransferase (ALT) and aspartate aminotransferase AST), the level of glutathione peroxidase (GSH-PX) and malondialdehyde (MDA) in liver tissue were observed. The pathological changes of liver tissue were observed by HE staining. Results: After acute liver injury induced by CCl4, ALT and AST levels in serum were significantly increased, while GSH-PX activity in liver tissue was significantly decreased and MDA content was significantly increased. Chinese Chaihu Guizhi Decoction - combined with decoction, Chaihu Guizhi Decoction - Xian Jian Houhe, Xiao Chai Hu Tang, Gui Zhi Tang can significantly reduce acute liver injury in rats serum ALT, AST activity and liver MDA content, improve liver GSH -PX activity. Conclusion: Chaihu Guizhi Decoction - combined with decoction, Chaihu Guizhi Decoction - Xianjianwuxia, Xiao Chai Hu Tang, Guizhi decoction can protect the liver from enzyme, anti-lipid peroxidation, acute liver injury caused by CCl4 in rats Has obvious protective effect. Chaihu Guizhi Tang - Xianzangzhongzhong in the interaction between Gui Zhi Tang and Xiao Chai Hu Tang may exist “evil” type “seven emotions ” relationship.
