采用200SW与0.1μg/ml10-羟基喜树碱单独或联合作用于人膀胱癌细胞(BT5637)。实验共分四组:对照组、HESW组、药物组和联合组。细胞集落形成率测定结果表明:HESW能增强10-羟基喜树碱对BT5637癌细胞生长抑制作用。3H-TdR掺入试验和FCMDNA含量测定结果证明HESW不仅增强10-羟基喜树碱抑制BT5637癌细胞DNA合成,而且亦可增强其抑制癌细胞分裂能力。由此,初步从细胞生物学水平阐明HESW增强抗癌药物抗肿瘤作用的机理
200 SW and 0.1 μg/ml 10-hydroxycamptothecin were used alone or in combination for human bladder cancer cells (BT5637). The experiment was divided into four groups: control group, HESW group, drug group and combination group. The results of colony formation assay showed that HESW could enhance the inhibitory effect of 10-hydroxycamptothecin on BT5637 cancer cells. The results of 3H-TdR incorporation assay and FCM DNA assay demonstrated that HESW not only enhanced 10-hydroxycamptothecin inhibition of DNA synthesis in BT5637 cancer cells, but also enhanced its ability to inhibit cancer cell division. Thus, the mechanism of the anti-tumor effect of HESW enhancing anticancer drugs was initially elucidated from the level of cell biology.
