长叶胡颓子不同生长部位体内外抗氧化活性及抗LDL氧化修饰的比较

目的:比较长叶胡颓子地上部位与地下部位体内外抗氧化作用,进行长叶胡颓子地上部位开发利用价值及抗动脉粥样硬化(AS)的可行性评价。方法:测定长叶胡颓子各部位在低、中、高质量浓度(5,10,20 g·L-1)对羟自由基(·OH)和超氧阴离子自由基(O-·2)的清除率;观察各部位在低、中、高质量浓度(5,10,20 g·L-1)对铜离子诱导的健康人静脉血低密度脂蛋白(LDL)氧化的影响,测定LDL氧化延迟时间(lag time)和达最大氧化速率时间(Tmax),硫代巴比妥酸(TBA)比色法测定反应液中丙二醛(MDA)含量;选用KM小鼠颈背部皮下注射5%D-半乳糖造模8周后,灌胃给予低、中、高剂量(2,5,10 g·kg-1)长叶胡颓子各部位提取液,14 d后眼眶取血,分离血清,观察长叶胡颓子各部位对血清总抗氧化能力(TAC)、谷胱甘肽过氧化物酶(GPX)、超氧化物歧化酶(SOD)等抗氧化酶活性、丙二醛(MDA)含量的影响。结果:长叶胡颓子地上部位低、中、高各剂量组对O-·2清除率分别为(58.26±6.14)%,(68.21±3.47)%和(74.67±4.10)%,对·OH清除率分别为(48.24±6.24)%,(78.21±7.24)%和(90.17±6.58)%,二者对自由基的清除率均高于地下部位各剂量组,同时长叶胡颓子地上部位和地下部位均能显著增加LDL的Lag time和Tmax,其结果与其减少铜离子诱导的MDA生成作用相一致;血清TAC,SOD,GPX活性均升高,能有效降低小鼠血清MDA含量,其地上部位活性升高显著。结论:长叶胡颓子各部位具有良好的体内外抗氧化活性,能明显提高血清及LDL抗氧化能力,同时长叶胡颓子地上部位抗氧化活性显著优于地下部位。 OBJECTIVE: To compare the antioxidation effect of Elaeagnus bockii and its underground parts both in vitro and in vivo, and to evaluate the feasibility of anti-atherosclerosis (AS) development and utilization of Elaeagnus bockii.
