【目的】探讨牛蒡子苷(arctiin)对小鼠3T3-L1前脂肪细胞增殖、分化的影响及其作用机制。【方法】采用不同浓度牛蒡子苷作用于3T3-L1前脂肪细胞,四甲基偶氮唑盐(MTT)法检测其对细胞增殖的影响;油红O染色及分光光度法分析脂肪细胞的分化程度;实时荧光定量PCR检测细胞过氧化物酶增殖物激活受体γ(peroxisome proliferators activatedreceptorγ,PPARγ)和CCAAT/增强子结合蛋白α(CCTTA enhancer binging proteinα,C/EBPα)的表达情况。【结果】牛蒡子苷对3T3-L1细胞增殖无显著性影响(P>0.05),但可显著抑制3T3-L1前脂肪细胞向成熟脂肪细胞分化(P<0.05或P<0.01),且呈一定剂量依赖关系;与空白对照组比较,牛蒡子苷可显著降低PPARγmRNA、C/EBPαmRNA表达水平(P<0.05或P<0.01)。【结论】牛蒡子苷能抑制3T3-L1前脂肪细胞的成脂分化,其机制可能与其能抑制脂肪细胞分化相关调控因子PPARγ和C/EBPα的表达有关。
【Objective】 To investigate the effect of arctiin on proliferation and differentiation of mouse 3T3-L1 preadipocytes and its mechanism. 【Method】 The effects of arctiin on 3T3-L1 preadipocytes and the effects of MTT on cell proliferation were analyzed. Oil red O staining and spectrophotometry were used to analyze the differentiation of adipocytes The expression of peroxisome proliferators activated receptor γ (PPARγ) and CCAAT / enhancer binding protein α (C / EBPα) were detected by real-time fluorescence quantitative PCR. [Results] Arctigenin had no significant effect on the proliferation of 3T3-L1 cells (P> 0.05), but significantly inhibited the differentiation of 3T3-L1 preadipocytes into mature adipocytes (P <0.05 or P <0.01) Compared with the blank control group, arctiin significantly reduced the expression of PPARγ mRNA and C / EBPα mRNA (P <0.05 or P <0.01). 【Conclusion】 Arctiin can inhibit the adipogenic differentiation of 3T3-L1 preadipocytes, which may be related to the inhibition of the expression of PPARγ and C / EBPα related to adipocyte differentiation.
