目的:探讨人端粒酶逆转录酶(hTERT)启动子调控Bax基因在端粒酶阳性的腺样囊性癌细胞-SACC-83中表达的特异性,以及Bax对SACC-83细胞增殖的影响。方法:脂质体介导pACTERT-Bax质粒载体转染SACC-83和端粒酶阴性的人胚肺成纤维细胞(HEL),通过RT-PCR检测Bax表达,MTT法检测Bax基因表达对细胞增殖的影响,流式细胞仪检测细胞凋亡率。结果:hTERT启动子调控Bax基因在SACC-83细胞中明显表达,对该肿瘤细胞的增殖有明显的抑制作用(相对活性58.90%),且诱导细胞凋亡比例增加(11.62%);而在HEL细胞中hTERT启动子未能诱导Bax基因表达,对该细胞的增殖和凋亡无明显影响。结论:hTERT启动子可以诱导Bax基因在SACC-83中特异性表达及诱导细胞凋亡,同时还能消除Bax基因对正常细胞的潜在毒性。
Objective: To investigate the specificity of human telomerase reverse transcriptase (hTERT) promoter in regulating the expression of Bax gene in telomerase-positive adenoid cystic carcinoma cell line-SACC-83 and the effect of Bax on the proliferation of SACC-83 cells . Methods: SACC-83 and human telomerase-negative human embryonic lung fibroblasts (HEL) were transfected with plasmid pACTERT-Bax by lipofectamine. The expression of Bax was detected by RT-PCR. The expression of Bax gene was detected by MTT assay. The effect of apoptosis was detected by flow cytometry. Results: Expression of Bax gene was significantly inhibited by hTERT promoter in SACC-83 cells, which significantly inhibited the proliferation of tumor cells (relative activity 58.90%) and induced apoptosis (11.62%). In HEL Cells hTERT promoter failed to induce Bax gene expression, the proliferation and apoptosis of the cells had no significant effect. Conclusion: The hTERT promoter can induce Bax gene expression in SACC-83 cells and induce apoptosis, and at the same time eliminate the potential toxicity of Bax gene to normal cells.
