不同浓度的石棉纤维与AM共同培养24h后,取上清液加入WI-38细胞培养,分别用3H-胸腺嘧啶(3H-TdR)掺入法和14C-羟脯氨酸(14-Pro)掺入法观察AM上清液对WI-38细胞增殖和胶原合成的影响,并用氯胺-T法测定成纤维细胞总羟脯氨酸含量。结果显示,石棉诱导的AM上清液能显著增加WI-38细胞增殖和胶原合成。3HTdR掺入的cpm达5205,14C-Pro掺入的cpm为7891,远高于对照组537和830(P<0.01)。经此AM上清液作用的WI-38细胞总羟脯氨酸含量为25.86μg/ml,显著高于TiO2对照组14.79μg/ml(P<0.01)。
After cultured with different concentrations of asbestos fibers for 24 h, the supernatant was added to WI-38 cells for culture. The 3H-thymidine incorporation and 14-hydroxyproline (14-Pro) The effects of AM supernatant on the proliferation and collagen synthesis of WI-38 cells were observed and the total hydroxyproline content in fibroblasts was determined by chloramine-T method. The results show that asbestos-induced AM supernatant can significantly increase WI-38 cell proliferation and collagen synthesis. The cpm of 3HTdR incorporation was 5205, and the cpm incorporation of C-C was 7891, much higher than that of control group (537 and 830) (P <0.01). The total hydroxyproline content of WI-38 cells treated with AM supernatant was 25.86μg / ml, which was significantly higher than that of the control group (14.79μg / ml) (P <0.01).
