采用斑马鱼表达谱芯片(Agilent,4×44 K)研究了稀有鮈鲫在鱼体代谢程度各异的物质雌二醇(E2)、二甲苯麝香(MX)及六氯苯(HCB)摄食暴露下不同时间点的代谢相关基因响应、表达趋势与特征,筛选出gla、ND4、psph以及gss等4个基因表达趋势与可代谢物质(MX及E2)鱼体代谢程度、较为一致的代谢相关基因。通过实时荧光定量PCR(q RT-PCR)验证4个基因的表达情况,进一步比较了相同浓度MX摄食暴露下,4个基因在稀有鮈鲫与斑马鱼中的表达情况。4个基因在2种鱼中的表达趋势与特征一致,其中gla、ND4及gss等3个基因,在物质暴露的早期阶段(3或4 d)出现1.5倍以上的上调,可能在早期揭示物质在鱼体的代谢能力,为简化或优化生物蓄积性测试及评估提供依据。
The feeding exposure of estradiol (E2), xylene musk (MX) and hexachlorobenzene (HCB) were studied by using zebrafish expression profiling chip (Agilent, 4 × 44 K) Metabolism related genes, expression trends and characteristics at different time points were screened. The expression trends of gla, ND4, psph and gss were screened and the metabolism of metabolites (MX and E2) . The expression of four genes was verified by real-time quantitative PCR (q RT-PCR), and the expression of four genes in rare crucian carp and zebrafish at the same concentration of MX was further compared. The expression pattern of four genes in the two species of fish showed the same trend. The three genes, gla, ND4 and gss, were up-regulated more than 1.5-fold in the early stages of substance exposure (3 or 4 days) The ability of the fish to metabolize provides a basis for simplifying or optimizing bioaccumulation testing and assessment.
