目的 :研究商陆素 -脾细胞条件培养基 (PWM-SCM)对小白鼠骨髓造血祖细胞的作用。方法 :用 micro-ELISA方法测定 PWM-SCM中 IL-2、IL-3、IL-4和 IL-6的含量 ,在 CFU-GM和CFU-E培养体系中加入 PWM-SCM,观察其对造血祖细胞的作用。结果 :PWM-SCM中 4种细胞因子的含量 :IL -2为 72 .5± 1 .1 pg/ml,IL -3为 43 3 .3± 1 1 .6pg/ml,IL -4为 2 8.9± 0 .4pg/ml,IL -6为 1 43 .3 pg/ml,其测定值均高于对照组 RPMI-1 64 0完全培养基中测定的含量 (P<0 .0 5~ 0 .0 1 )。骨髓造血祖细胞培养显示 ,PWM-SCM对 CFU-GM无影响 ,但 CFU-E的集落数由 6.2± 1 .4增加到 3 4.0± 2 .5 ,有非常显著性差异 (P<0 .0 0 1 )。结论 :PWM-SCM中含有多种细胞因子 ,对小白鼠骨髓造血祖细胞有集落刺激活性 ,对 CFU-E的形成有协同促进作用。
AIM: To investigate the effects of melatonin-splenocyte conditioned medium (PWM-SCM) on bone marrow hematopoietic progenitor cells of mice. Methods: The levels of IL-2, IL-3, IL-4 and IL-6 in PWM-SCM were determined by micro-ELISA. PWM-SCM was added into CFU-GM and CFU- The role of progenitor cells. Conclusion: There are many kinds of cytokines in PWM-SCM, which have colony-stimulating activity on mouse bone marrow hematopoietic progenitor cells and promote the formation of CFU-E.
