目的 探索鸟氨酸脱羧酶抗酶抑制子基因在小鼠发育过程中的功能.方法 用特殊的基因诱捕载体(gene trapping vector)制作了鸟氨酸脱羧酶抗酶抑制子基因敲除小鼠.用Western印迹、RT-PCR、X-gal染色方法分析该基因在突变小鼠体内的表达.PCR法鉴定小鼠不同发育时期的基因型.结果 鸟氨酸脱羧酶抗酶抑制子基因在纯合型突变小鼠体内被抑制后,纯合型突变小鼠在出生后死亡.结论 鸟氨酸脱羧酶抗酶抑制子基因是致死基因,对小鼠生存有不可缺少的作用.
