目的探讨慢性粒细胞白血病(CML)患者骨髓单个核细胞(BMMNC)中富半胱氨酸61基因(Cyr61)的表达水平及其临床意义。方法应用半定量RT-PCR和免疫组化技术分别检测38例CML患者和10例正常对照者(正常对照组)骨髓单个核细胞中Cyr61mRNA和蛋白水平的表达情况,CML患者中慢性期20例(慢性期组),加速期11例(加速期组),急变期7例(急变期组)。结果38例CML患者中,慢性期组、加速期组、急变期组Cyr61mRNA的表达明显高于正常对照组(P<0.05)。Cyr61基因在加速期及急变期的表达高于慢性期(P<0.05),Cyr61基因在加速期和急变期的表达虽升高,但两者差异无统计学意义(P>0.05)。Cyr61蛋白在CML中的表达明显高于正常对照组,CML加速期和急变期中Cyr61蛋白的表达高于慢性期(P均<0.05)。结论Cyr61基因参与了CML的发生、发展过程,且与CML的急变有关,可作为预示CML急变的实验依据。
Objective To investigate the expression of Cyr61 in bone marrow mononuclear cells (BMMNC) of patients with chronic myeloid leukemia (CML) and its clinical significance. Methods The expression of Cyr61 mRNA and protein in bone marrow mononuclear cells of 38 patients with CML and 10 normal controls (normal control group) were detected by semi-quantitative RT-PCR and immunohistochemistry. Among the 20 patients with chronic myelogenous leukemia (CML), 20 Chronic phase group), 11 cases of acceleration phase (accelerated phase group), 7 cases of acute phase (blastic phase group). Results The expression of Cyr61mRNA in chronic phase group, accelerated phase group and acute phase group were significantly higher than those in normal control group (P <0.05). The expression of Cyr61 was significantly higher in the accelerated phase and the blastic phase than in the chronic phase (P <0.05). The expression of Cyr61 in the accelerated phase and the blastic phase was not significantly different (P> 0.05). Cyr61 protein expression in CML was significantly higher than the normal control group, CML accelerated phase and blast crisis Cyr61 protein expression was higher than the chronic phase (P all <0.05). Conclusion The Cyr61 gene is involved in the pathogenesis and development of CML, and is related to the rapid change of CML, which may be used as an experimental basis for predicting the rapid change of CML.
