目的观察胆汁对离体血管平滑肌细胞(SMC)的作用,探索经颈静脉肝内门腔静脉分流术后狭窄的形成原因。方法用离体细胞培养法,将平滑肌细胞放入有不同浓度胆汁的培养液中培养。在3、10、14d收获细胞,每一收获点中每组为6个样本。检测脱氧核糖核酸(DNA)和总蛋白含量。结果此前预试验2.5%、5.0%、10.0%胆汁培养基,SMC在3d内全部死亡。离体细胞培养(1.0%胆汁)实验组中的DNA和总蛋白量明显较对照组少(P<0.05),且随培养时间延长其差异增加。结论2.5%~10.0%胆汁能杀死SMC,1.0%胆汁可抑制SMC生长。
Objective To observe the effect of bile on isolated vascular smooth muscle cells (SMC) and to explore the causes of stenosis after transjugular intrahepatic portosystemic shunt. Methods In vitro cell culture method, the smooth muscle cells into culture medium with different concentrations of bile culture. Cells were harvested at 3, 10, 14 days with 6 samples per group for each harvest point. Deoxyribonucleic acid (DNA) and total protein were detected. Results Previously, 2.5%, 5.0% and 10.0% of bile culture media were pre-tested and SMCs all died within 3 days. The amount of DNA and total protein in the experimental group of ex vivo cell culture (1.0% bile) was significantly less than that of the control group (P <0.05), and the difference was increased with the incubation time. Conclusions 2.5% ~ 10.0% bile can kill SMC. 1.0% bile inhibits SMC growth.
