[目的]了解孕晚期顺铂暴露是否会诱导子代豚鼠耳蜗毛细胞损伤。[方法]将孕晚期豚鼠12只随机分为两组,在孕50 d至56 d,顺铂组经腹膜腔注射顺铂1.5 mg/kg.d,对照组注射生理盐水1.5 mg/kg.d。出生后14d,应用原位末端转移酶标记技术(TUNEL)和Caspase-3免疫组化染色检测毛细胞凋亡情况。[结果]孕晚期顺铂暴露的子代豚鼠耳蜗毛细胞有凋亡发生及Caspase-3蛋白表达。[结论]孕期顺铂暴露可诱导子代豚鼠耳蜗毛细胞Caspase-3表达及凋亡发生。
[Objective] To understand whether exposure of cisplatin in the third trimester of pregnancy will induce hair cell damage of guinea pig cochlea. [Method] Twelve guinea pigs in the third trimester were randomly divided into two groups. At 50 d to 56 d of pregnancy, the cisplatin group was injected intraperitoneally with cisplatin 1.5 mg / kg.d and the control group with normal saline 1.5 mg / kg.d . 14 days after birth, hair cell apoptosis was detected by TUNEL and Caspase-3 immunohistochemical staining. [Results] The apoptosis of cochlear hair cells and the expression of Caspase-3 protein in cydia of guinea pigs exposed to cisplatin in the third trimester of pregnancy. [Conclusion] Cisplatin exposure during pregnancy can induce caspase-3 expression and apoptosis in cochlear hair cells of guinea pigs.
