采用ELISA方法检测了30例癫痫患者外周血单个核细胞(PBMC)经脂多糖(LPS)诱生后培养上清中的细胞因子水平。结果发现,癫痫患者PBMC分泌肿瘤坏死因子(TNF-α)和白细胞介素6(IL-6)的水平分别为(5.705±0.702)μg/L和(177.60±54.50)ng/L,较正常对照组[分别为(3.118±0.373)μg/L和(78.67±6.47)ng/L]明显升高(均为P<0.01),并且TNF-α与IL-6的升高存在明显的正相关(r=0.705,P<0.05)。应用Dot-ELISA及图像分析技术证实,癫痫患者PBMC表面人白细胞Ⅱ类抗原(HLA-Ⅱ抗原)的表达密度也显著高于正常对照组(P<0.01)。结果提示,IL-6、TNF-α的分泌紊乱及HLA-Ⅱ类抗原的表达异常可能与癫痫的发病有关。
ELISA was used to detect the level of cytokines in the culture supernatant of peripheral blood mononuclear cells (PBMCs) induced by lipopolysaccharide (LPS) in 30 patients with epilepsy. The results showed that the levels of tumor necrosis factor (TNF-α) and interleukin-6 (IL-6) secreted by PBMCs in epileptic patients were (5.705 ± 0.702) μg / L and (177.60 ± 54.50 ) Ng / L were significantly higher than those in the normal control group [(3.118 ± 0.373) μg / L and (78.67 ± 6.47) ng / L, respectively] There was a significant positive correlation between TNF-α and IL-6 (r = 0.705, P <0.05). Dot-ELISA and image analysis confirmed that the expression level of human leukocyte antigen (HLA-Ⅱ) on the surface of PBMC in epileptic patients was also significantly higher than that in the normal control group (P <0.01). The results suggest that, IL-6, TNF-α secretion disorder and abnormal expression of HLA-Ⅱ antigen may be related to the pathogenesis of epilepsy.
