研究大鼠放射性皮肤溃疡组织中p53;MDM2表达。采用140只Wistar大鼠进行局部照射制备放射性皮肤溃疡动物模型,观察病变一年,然后采用p53(Goat),MDM2(兔)多克隆抗体和SP免疫组化方法检测皮肤溃疡组织中p53,MDM2的表达情况。结果表明;p53, MDM2的高表达阳性率分别为 9.7%(7/72)、 19.4%(14/72),两者阳性部位主要见于增生肥大的鳞状上皮细胞核、间质成纤维细胞及血管内皮细胞核。p53,MDM2基因的变化及蛋白的高表达可能与放射性皮肤溃疡癌变及不愈合有关。
To investigate the expression of p53 and MDM2 in rat skin with radioactive skin ulcers. One hundred and forty Wistar rats were used for local irradiation to establish a model of radiation skin ulcer. The lesions were observed for one year. Then p53, MDM2 (rabbit) polyclonal antibody and SP immunohistochemical method were used to detect the expression of p53, MDM2 Express the situation. The results showed that the positive expression rates of p53 and MDM2 were 9.7% (7/72) and 19.4% (14/72), respectively. The positive expression of p53 and MDM2 was mainly found in the hypertrophy of squamous cell nucleus and mesenchyme Fibroblast and vascular endothelial cell nucleus. The changes of p53 and MDM2 genes and the high expression of proteins may be related to the carcinogenesis and non-union of radios skin ulcers.
