利用ISSR标记,对来源于中国7个省(市)的44个扁蓿豆居群的亲缘关系进行了分析。从80条ISSR引物中筛选出多态性强、重复性好的16条引物,对44个扁蓿豆居群基因组DNA进行扩增,共扩增出133条谱带,平均每个引物扩增出8.31条带,其中多态性带115条,多态性位点百分率为87.08%。扁蓿豆种质间遗传相似系数变化范围在0.436~0.908之间。利用UPGMA聚类分析,44个扁蓿豆居群被划分为8个类群,与地理来源有较高的相关性。
The ISSR markers were used to analyze the genetic relationship among 44 populations of Medicago from the seven provinces (cities) in China. Sixteen primers with strong polymorphism and good repeatability were screened from 80 ISSR primers. Genomic DNA of 44 alfalfa populations was amplified and a total of 133 bands were amplified. The average amplification of each primer There were 8.31 bands, of which there were 115 polymorphic bands with a percentage of polymorphic loci of 87.08%. The genetic similarity coefficient of Medicago luteosana germplasm ranged from 0.436 to 0.908. Using UPGMA cluster analysis, 44 populations of Medicago rizedatus were divided into 8 groups, which had a high correlation with geographical origin.
