目的:观察外源性RASSF1A基因对食管癌细胞的增殖作用并探讨机制。方法:将包含RASSF1A基因的质粒转染食管癌细胞EC9706,建立稳定转染细胞克隆,Western blot检测RASSF1A基因表达,通过细胞生长曲线检测细胞生长活性和增殖能力、流式细胞仪检测对细胞周期的影响、裸鼠移植瘤实验检测转染细胞的体内成瘤特性。结果:稳定表达RASSF1A基因的EC9706细胞中RASSF1A蛋白表达增加;细胞生长速度明显减慢;细胞周期中G1/G0期比例明显增加,S期比例减少;EC9706细胞的裸鼠致瘤能力被抑制。结论:RASSF1A基因能显著抑制食管癌细胞在体内外的生长,其机制可能与该基因诱导凋亡、抑制增殖作用有关。
Objective: To observe the effect of exogenous RASSF1A gene on the proliferation of esophageal cancer cells and explore the mechanism. METHODS: A plasmid containing RASSF1A gene was transfected into esophageal cancer EC9706 cells to establish a stably transfected cell clone. RASSF1A gene expression was detected by Western blot. The cell growth activity and proliferation ability were measured by cell growth curve. The cell cycle was detected by flow cytometry. Effects of tumor xenografts in nude mice The in vivo tumorigenicity of transfected cells was examined. RESULTS: The expression of RASSF1A protein in EC9706 cells stably expressing RASSF1A gene increased; the growth rate of the cells was significantly slower; the proportion of G1/G0 phase in the cell cycle increased significantly, and the proportion of S phase decreased; the tumorigenicity of EC9706 cells in nude mice was inhibited. Conclusion: RASSF1A gene can significantly inhibit the growth of esophageal cancer cells in vitro and in vivo, and its mechanism may be related to the induction of apoptosis and inhibition of proliferation of this gene.
