大肠杆菌不耐热肠毒素(LT)是一种有效的粘膜免疫原,口服或鼻内接种后可诱导高分泌性和全身性抗体应答。此外,LT在抗其他共服抗原的抗体应答中起到佐剂作用。为了研究LT亚单位的粘膜免疫原性和佐剂作用,作者从超量产生的大肠杆菌培养基中分别克隆和提纯LT全毒素和无毒性B亚单位(LTB),并分别用重组 LT、LTB及两者混合物鼻内免疫小鼠,检测它们诱导血清LTB特异性IgG、IgA和粘膜S-IgA的能力。
Escherichia coli Heat-labile Enterotoxin (LT) is a potent mucosal immunogen that induces both high-secretion and systemic antibody responses after oral or intranasal inoculation. In addition, LT acts as an adjuvant against other antibody responses that co-antigen. To investigate the mucosal immunogenicity and adjuvant effect of LT subunit, the authors cloned and purified LT holotoxin and non-toxic B subunit (LTB) respectively from overproducing Escherichia coli medium, and used the recombinant LT, LTB and Mice were immunized intranasally with a mixture of the two to test their ability to induce LTB-specific IgG, IgA, and mucosal S-IgA.
