目的本研究在大鼠Walker256实体瘤射频消融术(RFA)后应用不同肿瘤抗原致敏的DC瘤苗,探讨其对大鼠抗肿瘤免疫的不同影响。方法38只荷Walker256实体瘤SD大鼠随机分成3组:对照组行RFA治疗,实验组1行RFA+冻融抗原致敏DC治疗,实验组2行RFA+热休克冻融抗原致敏DC治疗。治疗前及治疗后第4天分别测量各组大鼠外周血中CD4+T淋巴细胞、CD8+T淋巴细胞及CD4+/CD8+比值,记录大鼠的荷瘤生存期。结果治疗后各组荷瘤大鼠外周血中CD8+T细胞均呈下降改变,其中,下降的程度为实验组2>实验组1>对照组(P<0.05);CD4+/CD8+比值的变化,实验组2的升高与其他两组相比差异有统计学意义(P<0.05);实验组1的升高与对照组比差异无统计学意义(P>0.05)。荷瘤生存期:实验组2>实验组1>对照组(P<0.05)。结论在RFA后应用不同抗原致敏的DC可不同地程度改善大鼠的抗肿瘤免疫,冻融热休克抗原致敏DC较未热休克冻融抗原致敏DC能更有效地激发荷瘤大鼠的抗肿瘤免疫机能。
Objective To study the different effects of different tumor antigen-primed DCs on rat antitumor immunity after rat Walker 256 solid tumor radiofrequency ablation (RFA). METHODS: Thirty-eight Walker 256 solid tumor-bearing SD rats were randomly divided into three groups: control group received RFA treatment, RFA + freeze-thaw antigen-sensitized DC group in experimental group, and RFA + heat shock freeze-thaw antigen-induced DC treatment in experimental group. The CD4 + T lymphocytes, CD8 + T lymphocytes and CD4 + / CD8 + ratio in peripheral blood of each group were measured before treatment and 4 days after treatment, and the tumor-bearing survival of the rats was recorded. Results After treatment, the levels of CD8 + T cells in the peripheral blood of all the tumor-bearing rats decreased in the experimental group 2> experimental group 1> control group (P <0.05); the ratio of CD4 + / CD8 + There was a significant difference between the experimental group 2 and the other two groups (P <0.05). There was no significant difference between the experimental group 1 and the control group (P> 0.05). Tumor survival: experimental group 2> experimental group 1> control group (P <0.05). Conclusions DCs sensitized with different antigens can improve antitumor immunity of rats to different extent after RFA. Cold-thawed heat-shocked antigen-induced DCs can stimulate tumor-bearing rats more effectively than DCs exposed to heat-shock Anti-tumor immune function.
